Preparation Method

Semaglutide is prepared using a combination of solid-phase and liquid-phase synthesis methods. The linear peptide Aib8, Arg34-GLP-1(7-37) and a fatty side-chain acylating agent are prepared via solid-phase synthesis; the linear peptide is then modified at the Lys26 residue using the synthesized fatty chain acylating agent under alkaline conditions to yield semaglutide. The biological activity of the prepared sample is subsequently determined using a time-resolved fluorescence resonance energy transfer (TR-FRET) immunoassay.
